Abstract :

In several contexts (medical, military or civil), it is necessary to identify and/or quantify rapidly a biological or chemical element present in very low quantity in a complex mixture.

Existing conventional analysis techniques suffer from a lack of precision and sensitivity especially when considering very low quantity of biological or chemical elements. The development of sensitive analytical techniques becomes a necessity to overcome this limitation.

Single-molecule sensing technique based on the electrical detection of single molecules through a nanometer-sized protein channel (biological nanopore) presents a higher precision and sensitivity than conventional techniques.

The aim of the thesis project is to use the technology of single-molecule electrical detection through a biological nanopore to develop an innovative, ultra-fast, low-cost, selective, and highly resolutive nanosensor allowing analysis of very low quantity of biological elements such as DNA or proteins. Upcoming applications concern high-throughput DNA and protein sequencing, size-discrimination of peptides, and analysis (identification and quantification) of very small amount of peptide impurities, with a single amino acid resolution.
